Recent Work on Skin Permeation and Skin Uptake

Abstract

The permeability of human skin from water is characterized by the permeability coefficient, kp. There are now a number of in silico calculations of kp, that have reasonable predictive power, so that it is not too difficult to predict kp from structure. Exceptions are strong acids and strong bases under ionizing conditions, but this problem has only recently been recognized. There is only limited data on water to skin partition, Ksc, but it is evident that neither log kp nor log Ksc are well modeled by any experimental process studied to date. In view of the unique character of the stratum corneum, which is the main rate-determining feature of skin, it is not surprising that no experimental models have been identified.
